Introduction of the Research Group

The BTRG research group is primarily concerned with the study of the in vitro ADME and physicochemical properties of active pharmaceutical agents, drug formulations, and development of related biomimetic models. Main methods belonging to our activity: - kinetic and thermodynamic solubility and dissolution tests - investigation of the solubility conditions of active substances, especially through the action of certain excipients (eg cyclodextrins) - pKa measurement by UV-pH titration method - logP measurement by equilibrium distribution and HPLC methods - in vitro tissue-specific (blood-brain barrier (BBB), gastrointestinal, cornea, nasal, liver and heart) lipid membrane permeability models: PAMPA and side-bi-side systems - HSA binding study by rapid equilibrium dialysis and specific HPLC system - chemical stability measurement in physiological buffer systems (PBS, SGF, SIF, Fassif, Fessif) - metabolic stability and intrinsic clearance study in the liver microsome system - CyP inhibition assay - production of an oxidative metabolite in a metalloporphyrin-based flow chemical system - investigation of biological oxidation transformations in electrochemical systems - development and testing of in silico ADME and PD models (supporting software: ACD / Percepta, Marvin Sketch)

Achievements

- Fine-tuning and development of tissue-specific PAMPA models: BBB, buccal, corneal, nasal
- Development of an in silico QSPR model for corneal permeability
- Application of PAMPA systems in dereplication of natural products from plants
- Investigation of the interaction between active substances and macromolecules based on changes in proton dissociation processes (pKa): drug-serum albumin and drug-cyclodextrin systems
- Synthesis and investigation of oxidative drug metabolites using continuous-flow biomimetic systems (liver-on-a-chip)
